There are few changes to the Mercedes-Benz R-Class for 2010. The 2009 R320 BlueTec is now known as the R350 BlueTec, yet it has the same 3.0L turbo-diesel engine as before. Style-wise, the 2010 R-Series features a slightly larger front grille, more squared-off headlights and new taillights. New exterior colors available are Palladium Silver and Steel Grey. Most notable for 2010 is the addition of Mercedes-Benz' 'Post-Safe' system, which unlocks the doors and shuts off the engine and fuel system after an accident. This feature complements Mercedes-Benz' PRE-SAFE system, which prepares the vehicle if a collision is anticipated. Both PRE-SAFE and 'Post-Safe' are now standard equipment.

A new, diesel-powered R320 BlueTec model joins the R-Class lineup for 2009; the model utilizes clean diesel technology to provide a highway driving range of more than 500 miles as well as impressive trailer-towing capabilities. Several technology features are new across the R-Class line for 2009. All models get a newly revised version of Mercedes-Benz' COMAND screen-driven interface system, which accesses climate control and audio functions. The PRE-SAFE system, which primes safety systems if a collision is anticipated, is now standard equipment. Also, 4MATIC all-wheel drive is now standard on both R-Class models.

Inspired by Mercedes-Benz's Vision GST concept design, the R-Class production car was developed during the Daimler Chrysler era in which it shared a platform with the Chrysler Pacifica. Featuring three rows of premium passenger seating, up to 85 cubic feet of storage came available inside the R-Class. For its 2006 debut, the Mercedes-Benz R-Class was available as the V-6 powered R350 or an R500 with a V-8 engine (The R500 was dropped after the 2007 model year). The second year of production for the R-Class consisted of two new trim variants. Mercedes-Benz provided the option of an R320 CDI diesel model as well as an R63 AMG. The high-performance 503-horsepower R63 AMG model existed for only the 2007 model year making it a rare version of the R-Class. In 2009, the diesel-engined R-Class incorporated a new, cleaner-operating 3.0-liter BlueTEC engine. All Mercedes-Benz R-Class vehicles sold in the United States were exclusively equipped with 4Matic all-wheel drive. Struggling to meet sales expectations, the R-Class was dropped after the 2011 model year.
